Immunohistochemistry Dilution: 1:1,000 - 1:5,000
Application Note: F(ab')2 Anti-Human IgG IgA IgM (H&L) Biotin Antibody has been assayed against 1.0 μg of Human IgG in a standard capture ELISA using Peroxidase Conjugated Streptavidin #S000-03 and ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) code # ABTS-100 as a substrate for 30 minutes at room temperature. A working dilution of 1:10,000 to 1:50,000 of the reconstitution concentration is suggested for this product. Suitable for immunomicroscopy and flow cytometry or FACS analysis as well as other antibody based fluorescent assays requiring lot-to-lot consistency.

Background: F(ab')2 Anti-Human IgG IgA IgM (H&L) Biotin Antibody generated in goat detects human (heavy and light chain) immunoglobulin G, A, and M. Immunoglobulin G binds to antigens and can neutralize or opsinize targets, and are predominantly involved in the secondary immune response. Immunoglobulin A (IgA) is an antibody that plays a critical role in mucosal immunity. IgA has two subclasses (IgA1 and IgA2) and can exist in a dimeric form called secretory IgA (sIgA). Immunoglobulin M, or IgM, is a pentamer composed of 5 immunoglobulin molecules linked through their F(c) domain by the J chain. Secondary Antibodies are available in a variety of formats and conjugate types. When choosing a secondary antibody product, consideration must be given to species and immunoglobulin specificity, conjugate type, fragment and chain specificity, level of cross-reactivity, and host-species source and fragment composition. F(ab')2 Antibody is ideal for investigators who routinely perform flow cytometry, immunohistochemistry or IHC and other immunoassays.
