Purified recombinant Human ALDH2 protein
Expression System: E.coli
Bioactivity: Specific activity is < 0.2 units/mL, and was obtained by measuring the increase of NADP in absorbance at 340 nm resulting from the reduction of NAD. One unit will oxidize 1.0 uM of acetaldehyde to acetic acid per minute at pH 8.0 at 25 °C in the presence of beta-NAD, potassium and thiols.
Molecular weight on SDS-PAGE will appear higher. - Purity

- ALDH2 protein has been used in SDS PAGE and may be suitable for use in other assays to be determined by the end user.
- Assay Procedure

- Prepare a 3 mL reaction mixture into a suitable container: The final concentrations are 103 mM Tris, 0.67 mM beta-NAD, 100 mM potassium chloride, 10 mM 2-mercaptoethanol, 2 mM acetaldehyde, 0.0007 % (w/v) BSA.
2. Equilibrate to 25 °C and monitor at A340 nm until value is constant using a spectrophotometer.
3. Add 5 µg of recombinant ALDH2 into reaction mixture and mix immediately.
4. Record the increase at A340 nm for 5 minutes

ALDH2 (Aldehyde dehydrogenase 2 family) belongs to the aldehyde dehydrogenase family which catalyze the chemical transformation from acetaldehyde to acetic acid and is the second enzyme of the major oxidative pathway of alcohol metabolism. There are two major liver isoforms of this enzyme, cytosolic and mitochondrial, and they can be also distinguished by their electrophoretic mobilities, kinetic properties, and subcellular localizations. Recombinant human ALDH2 protein was expressed in E. coli and purified by using conventional chromatography techniques.
