First Metanephrine (Metadrenaline) and Normetanephrine (Normetadrenaline) are acylated. The subsequent competitive ELISA kit uses the microtiter plate format. The antigen is bound to the solid phase of the microtiter plate. The acylated standards, controls and samples and the solid phase bound analytes compete for a fixed number of antiserum binding sites. After the system is in equilibrium, free antigen and free antigenantiserum complexes are removed by washing. The antibody bound to the solid phase is detected by an antirabbit IgGperoxidase conjugate using TMB as a substrate. The reaction is monitored at 450 nm. Determination of unknown samples is achieved by comparing their absorbance with a reference curve prepared with known standard concentrations. During the sample preparation Metanephrine (Metadrenaline) is acylated.

Metanephrine is a metabolite of epinephrine created by action of catechol-O-methyl transferase on epinephrine. An article in the Journal of the American Medical Association, 2002, indicated that the measurement of plasma free metanephrines is the best tool in the diagnosis of pheochromocytoma, an adrenal medullary neoplasm. Normetanephrine is a metabolite of norepinephrine created by action of catechol-O-methyl transferase on norepinephrine. It is excreted in the urine and found in certain tissues. It is a marker for catecholamine-secreting tumors such as pheochromocytoma.
