The Total Bcl2 Enzyme ImmunometricAssay (EIA) kit is a complete kit for the determination of Bcl2 in cell lysates. Please read the complete kit insert before performing this assay. The kit uses a monoclonal antibody to human Bcl2 immobilized on a microtiter plate to bind the Bcl2 in the standards or sample. A recombinant human Bcl2 Standard is provided in the kit. After a short incubation, the excess sample or standard is washed out and a biotinylatedmonoclonal antibody to Bcl2 is added. This antibody binds to the Bcl2 captured on the plate. After a short incubation, the excess antibody is washed out and streptavidin conjugated to Horseradish peroxidase is added, which binds to the biotinylated monoclonal Bcl2 antibody. Excess conjugate is washed out and substrate is added. After a short incubation, the enzyme reaction is stopped and the color generated is read at 450 nm. The measured optical density is directly proportional to the concentration of Bcl2 in either standards or samples.

Bcl2 initiates a new gene family involved in the regulation of cell death and survival without affecting cell proliferation. This precise regulation and maintenance of balance between cell proliferation and cell death in multicellular organisms is critical for tissue homeostasis. Bcl2, encoded by a protooncogene, is an intracellular membrane-associated protein that functions to block programmed cell death. Expression of Bcl2 has been reported in a wide range of hematopoietic cells, non-neoplastic epithelia and epithelial malignancies. Bcl2 levels are low or undetectable in endothelial cells. The Bcl2 gene has a unique function among mammalian oncogenes as a negative regulator of apoptosis. It was first discovered because of its involvement in chromosomal translocations commonly found in lymphomas. Bcl2 is furthermore associated with stem cells committed to differentiation and morphogenesis.
