    Assay Procedure

    This mouse IL4 Enzyme Immunometric Assay (EIA) kit is a complete kit for the determintation of mouse IL4 in biological fluids. Please read the complete kit insert before performing this assay. The kit uses a monoclonal antibody to mouse IL4 immobilized on a microtiter plate to bind themouse IL4 in the standards or sample. A recombinant mouse IL4 Standard is provided in the kit. After a short incubation the excess sample or standard is washed out and abiotinylated monoclonal antibody to mouse IL4 is added. This antibody binds to the mouse IL4 captured on the plate. After a short incubation the excess antibody is washed out and Streptavidin conjugated to Horseradish peroxidase is added, which binds to the biotinylated mouse IL4 antibody. Excess conjugate is washed out and substrate is added. After a short incubation, the enzyme reaction is stopped and the color generated is read at 450 nm. The measured optical density is directly proportional to the concentration ofmouse IL4 in either standards or samples.

    Background
    Mouse Interleukin 4 (IL4) is an antiinflammatory glycoprotein of 120 amino acids with a molecular mass of 13.5 kDa. It has also been referred to as B cell growth factor (BCGF), B cell stimulating factor (BSF), mast cell growth factor, and T cell growth factor (TCFG)II. IL4 is a costimulator of B-cell proliferation, as well as a growth factor for T cells. IL4 regulates cytokine production through T, B, and NK cells, as well as monocytes, macrophages, fibroblast, and endothelial cells. Its inhibitory effects on cytokine production make it a potential candidate for use in chronic inflammatory disease. There is also a prospect that IL4 may play a role in antitumor activity. There is a 50 % amino acid level homology shared by human and mouse IL4.
