- Useful in Sandwich ELISA for Quantitative Detection of Antigen. Aliquot 0.1 mL per well of the 6000pg/mL, 3000pg/mL, 1500pg/mL, 750pg/mL, 419pg/mL, 187.5pg/mL, 93.7pg/m human IL-18BP standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of human cell culture supernates, serum, plasma(heparin, EDTA) or urine to each empty well. It is recommended that each human IL-18BP standard solution and each sample be measured in duplicate.

Background: Interleukin-18-binding protein, also known as IL18BPa, is a protein that in humans is encoded by the IL18BP gene. It is mapped to 11q13.4. The protein encoded by this gene is an inhibitor of the proinflammatory cytokine IL18. This protein binds to IL18, prevents the binding of IL18 to its receptor, and thus inhibits IL18-induced IFN-gamma production. This protein is constitutively expressed and secreted in mononuclear cells. The expression of this protein can be enhanced by IFN-gamma. It has been found that IL18BP expression in intestinal tissue is increased in endothelial cells as well as cells of the submucosa and overlying lymphoid aggregates in Crohn disease.
