Tissue Specificity: Expressed at high levels in the heart, skeletal muscle, testis and brain (at protein level). In the brain, detected in the cerebral cortex neurons through layers II to VI. In the hippocampus, detected in the CA1 to CA3 pyramidal regions and in the granule and polymorph layers of dentate gyrus. Weak expression in the striatum. In substantia nigra, detected in solitary cells that did not express tyrosine hydroxylase, a marker for dopaminergic neurons. Relatively high expression in the Purkinje cells of the cerebellum and in regions of the brain stem, including the locus coeruleus.
Background: Cerebral dopamine neurotrophic factor also known as ARMET-like protein 1 or is a protein that in humans that is encoded by the CDNF gene. CDNF and MANF are secreted proteins with 8 conserved cysteine residues, which predict a unique protein fold and define an evolutionarily conserved protein family. The human CDNF mRNA encodes a 187-amino acid protein with a predicted signal peptide and apparently no prosequence. The predicted secondary structure, like that of MANF, is dominated by alpha-helices. Human CDNF shares 59 % amino acid identity with human MANF, and 49 % and 46 % identity with D. melanogaster and C. elegans Manf proteins, respectively. RT-PCR of mouse brain detected Cdnf transcripts in embryonic and postnatal stages, and in adult brain regions, including the striatum. In the midbrain, Cdnf mRNA was expressed from embryonic stages to the adult. High levels of Cdnf mRNA were observed in adult mouse heart, skeletal muscle, and testis. In agreement with the results from mouse, wide expression of CDNF transcripts was detected in human brain and nonneuronal tissues. Interestingly, levels of CDNF transcripts were relatively high in the corpus callosum and optic nerve, which are devoid of neuronal cell bodies and contain mostly axonal projections and oligodendrocytes. At postnatal stage P10 of the mouse brain, Cdnf staining was most intense in the hippocampus and thalamus. Cdnf signal was also detected in striatum and substantia nigra.
