MeDIP
-
- Application
- Methylated DNA Immunoprecipitation (MeDIP)
- Specificity
- The MeDIP Kit is tested for specific enrichment of 5-methylcytosine DNA from the remaining genomic DNA.
- Characteristics
- The MeDIP Kit is designed to immunoprecipitate and enrich for single-stranded DNA fragments containing 5-methylcytosine (5-mC). The MeDIP Kit contains a monoclonal 5-methylcytidine antibody, a bridging antibody and the necessary buffers to perform methylated DNA immunoprecipitation (MeDIP). The affinity of the antibody enables separation of 5-mC DNA from 5-hmC DNA using single-stranded input DNA.

- Comment
-
The MeDIP kit is run in duplicate on 500 ng of the provided MseI digested human genomic DNA in the presence of bridging antibody with either the 5-methylcytidine mouse monoclonal antibody or a negative control mouse IgG. The samples are immunoprecipitated according to the protocol listed in the manual. Following elution, the DNA is cleaned up using 's Chromatin IP DNA purification kit and assayed in real time PCR with the ZC3H13 PCR primer mix included in the MeDIP kit. The samples with the 5-methylcytidine antibody will show enrichment with the ZC3H13 locus, while samples immunoprecipitated with the mouse IgG should show no enrichment (Figure 1).
