    Sequence similarities: Belongs to the Toll-like receptor family.

    Tissue Specificity: Expressed at high level in placenta and pancreas. Also detected in CD11c+ immature dendritic cells. Only expressed in dendritic cells and not in other leukocytes, including monocyte precursors. TLR3 is the TLR that is expressed most strongly in the brain, especially in astrocytes, glia, and neurons. .

    human TLR3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for TLR3 has been precoated onto 96-well plates. Standards (NSO,K27-S711) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for TLR3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human TLR3 amount of sample captured in plate.
    Assay Procedure

    Aliquot 0.1 mL per well of the 10000pg/mL, 5000pg/mL, 2500pg/mL, 1250pg/mL, 625pg/mL, 312pg/mL, 156pg/mL human TLR3 standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of human cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each human TLR3 standard solution and each sample be measured in duplicate.

    Background

    Protein Function: Key component of innate and adaptive immunity. TLRs (Toll-like receptors) control host immune response against pathogens through recognition of molecular patterns specific to microorganisms. TLR3 is a nucleotide-sensing TLR which is activated by double-stranded RNA, a sign of viral infection. Acts via the adapter TRIF/TICAM1, leading to NF-kappa-B activation, IRF3 nuclear translocation, cytokine secretion and the inflammatory response. .

    Background: Toll-like receptor 3(TLR3) also known as CD283(cluster of differentiation 283) is a protein that in humans is encoded by the TLR3 gene. The TLR3 gene was mapped to chromosome 4q35 by fluorescence in situ hybridization. TLR3 specifically recognizes double-stranded RNA(dsRNA) and induces multiple intracellular events responsible for innate antiviral immunity against a number of viral infections.
