Neutralization of human IL-23 bioactivity. It is recommended to establish the optimal antibody concentration for the assay system used. The antibody is also suitable for immunoassays. Neutralization Neutralization assay: Neutralization of human IL-23 induced IFN-γ production (see graph). To determine the neutralizing capacity and ND50, mixtures of various concentrations of mAb MT155, human recombinant IL-23 (10 ng/mL) and recombinant human IL-18 (50 ng/mL) were added to human PBMC. Following overnight incubation, cell supernatants were quantified in IFN-γ ELISA. Neutralization dose 50 (ND50): MT155 neutralized the effect of IL-23 at a ND50value of 0.2-0.8 μg/mL when 10 ng/mL recombinant human IL-23 was present. ND50 is defined as the concentration of antibody that results in 50 % inhibition of the biological response under the experimental conditions specified.
