Characteristics: Monoclonal Anti-Mouse CD45RB is recommended for use in flow cytometry for identification of CD45RB, an isoform of CD45. The C363-16A react with an isoform of CD45 with exon 5 splicing (encodes B determinant). It is a 220 kD glycoprotein expressed on peripheral B cells, naïve T cells, thymocytes, weakly on macrophages, and dendritic cells. It plays a critical role in TCR and BCR signaling. As T cells become activated and progress from naïve to memory cells, CD45RB expression is downregulated. Additionally, functionally distinct CD4+ T cell subsets, which secrete differing cytokine profiles, can be separated by CD45RB intensity. The primary ligands for CD45 are galectin-1, CD2, CD3, CD4 and Thy-1. The anti-mouse CD45RB monoclonal antibody reacts with the CD45 isoform containing the exon B dependent epitope. CD45RB is highly expressed on peripheral B cells, cytotoxic T cells, a subset of T helper cells and most thymoctyes.

Application Details

Application Notes: It is recommended for use in flow cytometry. This reagent is effective for direct immunofluorescence staining of human tissue for flow cytometric analysis. For immunofluorescent staining, the suggested use of this reagent is less or equal to 1 µg per 10^6 cells in 100 µL volume.

Background: CD45 is a family of single chain transmembraneous glycoproteins consisting of at least four isoforms which share a common large intracellular domain. Their extracellular domains are heavily glycosylated. The different isoforms are produced by alternative messenger RNA splicing of three exons of a single gene on chromosome 1. CD45 is expressed on cells of the human hematopoietic lineage with the exception of mature red cells. It is not detected on differentiated cells of other tissues. It is likely that CD45 plays an important role in signal transduction, inhibition or upregulation of various immunological functions.
