ELISA: Direct: To detect Human BD-3 by direct ELISA (using 100 μL/well antibody solution) thisantibody can be used at a concentration of 1.0 μg/mL. Used in conjunction with compatiblesecondary reagents, allows the detection of at least 0.2-0.4 ng/well of recombinant HumanBD-3. Sandwich: To detect Human BD-3 by Sandwich ELISA (using 100 μL/well antibody solution)this antibody can be used at a concentration of 0.25-1.0 μg/mL. This biotinylated antibody,used in conjunction with polyclonal anti-human BD-3, allows the detection of at least0.2-0.4 ng/well of recombinant Human BD-3. Western blot: To detect Human BD-3 by Western Blot analysis this antibody can be used ata concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hBD-3 is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Beta Defensin 3 exhibits antimicrobial activity against Gram-positive bacteria S.aureus and S.pyogenes, Gram-negative bacteria P.aeruginosa and E.coli and the yeast C.albicans. It kills multiresistant S.aureus and vancomycin-resistant E.faecium, but it appears to have no significant hemolytic activity. Beta Defensin 3 is highly expressed in skin and tonsils, and to a lesser extent in trachea, uterus, kidney, thymus, adenoid, pharynx and tongue. Low expression is observed in salivary gland, bone marrow, colon, stomach, polyp and larynx, with no expression in small intestine. Expression is induced by infection of bacteria and by interferon gamma.
