    Flow Cytometry (FACS), Immunofluorescence (IF)
    Characteristics
The A20 antibody reacts with CD45 (Leukocyte Common Antigen) on all leukocytes of mouse strains expressing the CD45.1 alloantigen (eg, RIII, SJL/J, STS/A, DA). This alloantigen was originally named Ly-5.2, and this was the designation at the time that the antibody was characterized. The designation was later changed from Ly-5.2 to Ly-5.1 to conform with the convention that the .2 alloantigen designations be assigned to the C57BL/6 strain. mAb A20 has been reported not to react with leukocytes of most other mouse strains, which express the CD45.2 alloantigen. Antibodies are supplied in buffer containing stabilizer and 0,05% sodium azide
    Immunogen
    SJL mouse thymocytes and splenocytes

    Isotype
    IgG2a
  • Application Notes
It is recommended for use in flow cytometry. The A20 antibody has been tested by flow cytometric analysis of mouse splenocyte suspensions. For immunofluorescent staining, the suggested use of this reagent is smaller than or equal to 1.0 µg per million cells in 100 µL volume. Cell number should be determined empirically but can range from 105to 108 cells/testIt is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

    Background
CD45 is a member of the Protein Tyrosine Phosphatase (PTP) family: Its intracellular (COOH- terminal) region contains two PTP catalytic domains, and the extracellular region is highly variable due to alternative splicing of exons 4, 5, and 6 (designated A, B, and C, respectively), plus differing levels of glycosylation. The CD45 isoforms detected in the mouse are cell type-, maturation-, and activation state- specific. The CD45 isoforms play complex roles in T-cell and B-cell antigen receptor signal transduction. The A20 antibody has been reported to inhibit some responses of B cells, from mice expressing the CD45.1 alloantigen, to antigens and LPS. CD45.1 is an alloantigen of CD45, expressed by Ly5.1 bearing mouse strains (e.g., RIII, SJL/J, STS/A, DA). CD45, a member of the protein tyrosine phosphatase (PTP) family, is a 180-240 kDa glycoprotein expressed on all hematopoietic cells except mature erythrocytes and platelets. There are multiple isoforms in the mouse that play key roles in TCR and BCR signal transduction. These isoforms are very specific to the activation and maturation states of the cell as well as specific cell type. The primary ligands for CD45 are galectin-1, CD2, CD3, CD4, TCR, CD22, and Thy-1. Structure of this protein is a Protein tyrosine phosphatase (PTP) family of 180-240 kDa that is distributed by all hematopoietic cells except mature erythrocytes and platelets of the CD45.1 strain of mice.
