Roundabout proteins 1-3, known as Robo1-3, are repulsive axon guidance receptors that bind to Slit, a repellent secreted by midline glia during development. Robo3 (also known as Rig1) is important for normal axon navigation at the ventral midline of the neural tube during development and is specifically expressed by commissural axons in rodents. Loss-of-function studies in mice show that Robo3 is required to allow commissural axons to enter the floor plate and cross to the contralateral side of the spinal cord.
