Specificity
The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific phosphopeptide. The antibody against non-phosphopeptide was removed by chromatography using non-phosphopeptide corresponding to the phosphorylation site. cdc25A (phospho-Ser75) antibody detects endogenous levels of cdc25A only when phosphorylated at serine 75.

Immunoaffinity chromatography.
Immunogen
The antiserum was produced against synthesized phosphopeptide derived from human cdc25A around the phosphorylation site of serine 75 (M-G-SP-S-E).
Suitable for use in Western blot (1: 500approx. 1: 1000) and Immunohistochemistry (1: 50approx. 1: 100). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Cyclin dependent kinases are positively regulated by association with cyclins and negatively regulated by binding to inhibitory subunits. The activity of cyclin dependent kinases is also regulated by the phosphorylation status, which is controlled by the antagonistic action of Wee1 kinase and CDC25 phosphatases. Three CDC25 genes are present in human cells: CDC25A, CDC25B, and CDC25C. These three genes function at different phases of the cell cycle.
